It takes quite a while to have a breed "breed true" several doggy generations at least. Probably more like 10-20 years.
That said, I like the idea but I'd say having them negitive on perception even if they are big is wrong. Dogs hearing, smell and even eyesight are more accute than people's by a long shot and so should really be getting a bonus to perception and initiative.
I would make the disadvantage that they have a natural propensity to fight other dogs, even other mastiffs, and that they were notorious barkers and would bark when attacking (training whith an anti bark collar has no effect as the dogs don't feel pain while in attack mode and surgery to retard the vocal cords is expesive and only ruduces the volume by 50%) which in turn bring more zed. Plus that they eat more like you said, and only have about a 1d4+4 year (natural) lifespan. Most usually only live about 4 years (2 years of training and 2 years of service)
